NPCs will “understand the state of the world” That will be a very long game in terms of content delivered over many years." The development is progressing well and is a beautiful world delivered by the Snowdrop engine. In a Q2 earnings call in 2022 (via TweakTown (opens in new tab)), Ubisoft CFO Frederick Duguet said, "Avatar Frontiers of Pandora is a big game next to the highly-awaited movie. Ubisoft is planning a long life for Avatar: Frontiers of Pandora when it releases later this year it seems. We just keep a close eye that they don’t do anything that’s not canonical in terms of Na’vi culture and what the RDA is doing on Pandora and all that sort of thing.” I don’t tell them what to do - they know their world, their business, their market. "We're very excited about what Ubisoft is doing with their game authoring. Confirming he's taking a hands-off approach, he explained: James Cameron revealed his thoughts on Ubisoft's upcoming game during an IGN (opens in new tab) interview in 2022. James Cameron shares his thoughts on Frontiers of Pandora The Na’vi jumps on a Great Leonopteryx, firing arrows at the helicopter, before a final arrow fixed with an explosive device hits the vehicle, which then bursts into flames. This trailer shows RDA helicopters on the warpath, trying to destroy a Na’vi. We’re hoping this could mean online play - we also hope we can customize our Na’vi.įrom the trailer, it also looks like Pandora’s native Na’vi will take on the RDA, like in the film, who threaten to destroy their world and have significant armed tech at their disposal. There also seems to be some sort of hub for Na’vi to congregate in. The world also seems to be inhabited by creatures we both have - and haven’t - seen in the film, so expect some surprises. We also see a Na’vi riding a mount - that doesn’t quite look like a Direhorse - suggesting that we will hopefully be able to partake in mounted combat, or we’ll at least be able to traverse Pandora on (kind of) horseback. The first-look trailer gives us a brief look at this first-person perspective. ![]() The description also states that the game will take players into a "standalone story" where they'll "play as a Na’vi and embark on a journey across the Western Frontier, a never-before-seen part of Pandora". ![]() The official site (opens in new tab) for the game describes it as a "first-person action adventure" that's set in an "immersive, open world". ![]() Details on Frontiers of Pandora’s story are a bit light currently, but we’ve been able to gather a few potential snippets from the reveal trailer and Ubisoft's site.

![]() What of the live-service focused Skull and Bones then? Game director Ryan Barnard promises that Ubisoft Singapore will be "communicating soon about our live plans" but reaffirms that Skull and Bones is indeed "a live game" and will receive the necessary support to back that title up. Whereas, Ghost Recon Breakpoint and The Division 2 have each received almost three years of support, while 2020's Assassin's Creed Valhalla received its most recent expansion earlier this year. If you look at something like Hyper Scape, the free-to-play battle royale from Ubisoft Montreal, its servers were taken offline fewer than two years after they were brought online. How long will Skull and Bones be free to sail the seas? It's a valid question, because Ubisoft has been a little inconsistent in this respect. "We intend to give new reasons for players to come back day-upon-day, and week-upon-week" So, should you solo players out there worry about being accosted by other pirates while playing? Barnard promises that "multiplayer is something you choose to do – either through uPlay with your friends or in-game, you can invite players to join your party." Oh, and Barnard confirms this piece of great news: "yes, Skull and Bones is fully crossplay across PS5, PC, Xbox Series X, Stadia, and Luna." Is Ubisoft committed to supporting Skull and Bones? You can definitely play alone, but part of the risk in our world is that, if you are the lone wolf, you could potentially become prey for other players." ![]() "We want players to have advantages when they group up and pirate together. That said, there are prominent pirates you will meet along the way, which will unveil a narrative arc – we want the players to be creating their own story," he says, before noting the importance of multiplayer. We are not a classic narrative-driven game – we are a live game, which we are committed to for years to come. So, what does the Skull and Bones of today offer the world? "You can play the game entirely solo," says game director Ryan Barnard, "however, we do look at everything with a multiplayer lens." When it was first unveiled to the public, Skull and Bones was set to be made up of two components: a single-player campaign, which would allow you to sail the Indian Ocean by yourself as a customizable pirate captain and a multiplayer experience, which would support five versus five players in tactical ship-to-ship combat across Disputed Waters. "Yes, Skull and Bones is fully crossplay across PS5, PC, and Xbox Series X" The cornerstone throughout the whole development of Skull and Bones is having best in class naval combat." Is Skull and Bones a multiplayer focused experience? So, where can Black Flag's lingering influence still be found most strongly through all of this? "The influence from Black Flag you will see in the naval combat, which is the common thread. Barnard says his key focuses are in ensuring that there's "depth to the naval combat" and "looking at the progression system with Infamy", as well as the "enemy factions you'll face" and "the depth of the crafting". "Since I joined, my focus has been about taking the creative vision to life," he says. "The team developed their expertise in Naval Combat through their co-development work so we decided to capitalize on that to create a totally new world – the world of Skull and Bones," says game director Ryan Barnard, who joined the project in June of last year after "the team had just passed a major internal milestone." It's worth remembering that Ubisoft delayed Skull and Bones into 2022 back in May 2021 – so whatever that milestone was, Ubisoft liked what it saw enough to allocate a year of additional resources behind the project. So, given that Skull and Bones has its developmental roots in a game that was released all the way back in 2013, how much of Assassin's Creed can be felt in the final release? ![]() Ubisoft Singapore, who collaborated with Ubisoft Montreal on the naval aspects of AC3, AC4, and Rogue, had initially set out to build a Black Flag expansion before its ambitions allegedly grew (opens in new tab). "The cornerstone throughout the whole development of Skull and Bones is having best in class naval combat"įamously, Skull and Bones started life after the world fell in love with the naval battles of Assassin's Creed 4: Black Flag. ![]() I wrote a function to do this, called CreateDepthOutline. We need to give all its pixels the same colour of 128 (ie roughly half the maximum of 255 for a colour), and give all the other pixels in the picture (ie all the pixels around the shark) a colour of 255 (because we want them to be the background). We want to put it in the middle distance, about halfway to the background (inside the image). But this picture needs to be redrawn as a “depth” image, that is, its pixels won’t contain colour, but information about how far away to put each pixel. Then, of course, you need the hidden picture. It needs to be quite noisy (lots of colour changes) to help make the hidden picture stand out. One is the pattern that you will see when you first look at the final image. If you want to know more about how stereoscopic pictures work, that’s a good place to go. The wider the gap between the pixels, the further away the pixel seems to be.įortunately, there are kind people who do all the hard work and share their code and explanations, and I have converted the code on this webpage to run in Codea. How it works, is that you match up pixels on the left and right of the picture at a certain distance from each other, so your left and right eye put them together and “see” the pixel at a certain distance away. Can you see the shark?Īnyway, this is how to create them with Codea.Ĭreating stereoscopic pictures is quite difficult. You can either see them or you can’t, usually. Still stuck on the gray Apple screen (which there’s never a loading bar, just a lone, gray Apple symbol). Eventually I found another thing to try: pressing down OPTION+COMMAND+R+P. I tried holding down the SHIFT button to start it in safe mode - a gray circle with a slash through it (□) popped up. So I researched, discovered the “white screen of death” and tried to resolve it based on what many sources say would fix a white screen. Note: each time it did this by itself and each I time I turned it off & on, there was no chime (or music or whatever it’s called - the loud noise a Mac makes when it comes on). White screen, clicking, Apple, black screen. The Apple would be there for a long time, and then the screen would go black, and then the whole process started over again. The screen stayed white for a long time, then eventually the gray Apple popped up but nothing else after that. It happened about 8 times and then stopped. I noticed a tapping/clicking noise coming from it. However, yesterday I woke it up, was about to start working on it when suddenly the screen went white. I know, it’s ancient - but it has been working great for a long time. ![]() Magic Beans are needed in order to sneak inside the Deku Palace's prison and talk to the Imprisoned Monkey, in effect learning the " Sonata of Awakening". Less sensitive than ELISA: Western blotting is less sensitive than ELISA, making it more difficult to detect low-abundance proteins. Ideal for identifying specific proteins: Western blots can be used to specifically identify the presence of a target protein in a sample, even when it is present at low levels or in a complex mixture. Longer process with more steps involved: Western blotting is a multi-step process that can take several hours (often overnight) to perform Using the Azure 400 allows you to simultaneously detect 3 target proteins image using three-color fluorescence. High-throughput: Due to being performed in a 96-well plate, more samples can be processed faster and with less sample needed. Since they are usually performed in 96-well plates, ELISAs allow for many conditions and repeat samples to be tested at once. Versatile: ELISA can be used for both qualitative and quantitative protein analysis on a wide variety of sample types, including serum, cell culture supernatants, and urine. Rapid and easy to perform: ELISA can be performed relatively quickly and requires minimal sample preparation, making it a convenient choice for routine assays. High background: ELISAs can have high background due to cross reactivity of primary and secondary antibodies. Quantitative analysis: ELISA can be used to measure the exact amount of target protein in a sample, making it a valuable tool for determining protein concentration or monitoring changes in protein levels over time. Limited multiplexing capability: Traditionally, only one protein can be targeted in each ELISA. This makes it an ideal choice for detecting rare or low-abundance proteins. High sensitivity: One of the major advantages of ELISA is its high sensitivity, making it capable of detecting very small amounts of target proteins, even in complex mixtures. They added these QconCAT standards to each lung tissue sample following extraction but prior to trypsin digestion. The team designed and synthesized two QconCAT genes that covered 56 lung-associated proteins (74 peptides), using transformed Escherichia coli for expression and subsequent purification of the SIL standards. used a l iquid chromatography (LC)-SRM approach in conjunction with stable isoptope labels (SIL) to characterize the proteomes. This involved tissue digestion in cyanogen bromide ( CNBr ) before standard trypsin digestion for mass spectrometry (MS) evaluation. The researchers used a standard protocol to create the acellular scaffolds but extended proteomic extraction procedures to capture insoluble proteins remaining in the ECM. The research team used lung preparations from adult Fischer rats, comparing ECM and cellular proteomes from decellularized tissue with those from untreated samples. However, preparing ECM samples for proteomic assessment is complicated, since the protein fraction of interest is frequently discarded as insoluble during standard preparation. Quantifying this following tissue processing would be a good step in evaluating decellularizing procedures. In order for host cells to establish successfully, sufficient ECM proteome must remain in situ to direct functionality. Decellularization processes can strip away more than just the cells from the underlying ECM, leaving a reduced environment for subsequent repopulation of functional cell s. These tissue scaffolds hold a great deal of promise however, problems remain with maintaining an optimal functional environment in balance with sufficient cell removal. This regenerative medicine option could minimiz e both immune rejection and loss of potential ly useful donor organs. One model under investigation involves recipient stem cells seeding decellularized tissue matrices from donor organs that may be too damaged to transplant whole. Alternatives in the future may eventually include a “grow your own” option, with in vitro generation of organs for transplant. Unfortunately, the laws of supply and demand rarely equalize for organ transplant, with many recipients spending months and years on a wait list until a donor arrives. 1 Using a quantitative approach, the researchers examined differences in protein abundance between normal lung tissue and acellular scaffolds to evaluate the effect of cell removal protocols that can affect residual functionality for in vitro organ generation. (2015) explore a targeted serial reaction monitoring (SRM) proteomics workflow that specifically addresses characterizing the extracellular matrix (ECM) proteome in decellularized rat lung scaffolds. |
